Computer designed moleculeThe chemical bond between carbon and fluorine is one of the strongest in nature, and has been both a blessing and a curse in the complex history of fluorocarbons. Now, in a powerful demonstration of the relatively new field of "computational chemistry," researchers at the National Institute of Standards and Technology (NIST) and the Interdisciplinary Network of Emerging Science and Technology group (INEST, sponsored by Philip Morris USA) have designed—in a computer—a wholly theoretical molecule to pull the fluorine out of fluorocarbons.

The potentially toxic components of the fluorocarbon are immobilized in the macrocycle until removed by a second reaction, an important feature for possible filtering systems.

At sea level, the strong C-F bond makes fluorocarbons thermally and chemically stable. As a result, fluorocarbons have been used in many commercial applications including refrigerants, pesticides and non-stick coatings. In the upper atmosphere, however, high-energy photons and highly reactive ozone molecules can break apart fluorocarbons, with the well-known consequence of a depleted ozone layer and increased ultraviolet radiation at ground level. A determined chemist can break down fluorocarbons at ground level with certain organometallic compounds, but the reactions take a long time at very high temperatures. Other known reagents are both highly toxic and inefficient, so chemists have been searching for an economical and environmentally friendly method to dispose of fluorocarbons.

Reasoning that the problem already may have been solved by nature, the NIST/Philip Morris team looked to an enzyme called fluoroacetate dehalogenase used by a South African bacterium, Burkholderia sp. The enzyme enables the bacterium to pull the fluoride ion out of sodium fluoroacetate (disrupting a poisonous compound) at room temperature and without problematic metal ions. Enzymes are giant molecules, evolved to survive and work in the complex environment of a living organism; they can be difficult and expensive to adapt to an industrial process. Instead, the research team applied basic quantum mechanical theory of electron structures in molecules, together with the example of a known molecule that binds to and extracts chlorine ions, to calculate the make-up and geometry of the critical "active site" in the enzyme that does the work. They then designed in software a large ring-shaped molecule to hold those components in just the right orientation to break the C-F bond in methyl fluoride, a simple fluorocarbon.

Researchers at the University of Texas now are synthesizing the new molecule to test its effectiveness. If it matches theoretical predictions, it will be the first example of a simple organic molecular system able to break C-F bonds without extreme temperature and pressure conditions, and a demonstration of a novel technique for designing man-made molecules that can mimic the extraordinary selectivity and chemical activity of natural enzymes. Notes lead researcher Carlos Gonzalez, "All of these useful things are in nature, you just have to find them and make them more efficient."

Learning how genes responsible for biomass breakdown are turned on could lead to better ethanol production Permalink

As the national push for alternative energy sources heats up, researchers at the University of Rochester have for the first time identified how genes responsible for biomass breakdown are turned on in a microorganism that produces valuable ethanol from materials like grass and cornstalks.

Waste products such as grass clippings and wood chips—once thought too difficult to turn into ethanol—may soon be fodder for hungry, gene-tweaked bacteria.

The findings in today's Proceedings of the National Academy of Sciences may empower scientists to engineer ethanol-producing super-organisms that can make clean-burning fuel from the nation's one billion unused tons of yearly biomass production.

"This is the first revelation of how a bacterium chooses from its more than 100 enzymes to break down a particular biomass," says David H. Wu, professor in the Department of Chemical Engineering at the University of Rochester. "Once we know how a bacterium targets a particular type of biomass, we should be able to boost that process to draw ethanol from biomass far more efficiently that we can today."

Ethanol holds the promise of a clean, renewable alternative to fossil fuels, but deriving it from plants is difficult. Producing it from corn is the easiest method, but doing so on a large scale would drive up the price of corn, corn starch, and even tangential foods like beef, since cows are fed on corn—not to mention all the energy spent fertilizing, maintaining, and harvesting a crop like corn. Conversely, deriving ethanol from plant materials such as the corn stalks and wood chips is challenging because the plants' cellulose is a very tough substance to break down, making for an inefficient process.

Wu's technique may prove much more effective than traditional methods. Instead of using separate steps to break down biomass into glucose and ferment the glucose into ethanol, as is currently done, Wu is working on a way to make a bacterium break down and ferment plant biomass efficiently in just one step.

Wu investigated C. thermocellum, which is a microorganism that has that ability to turn biomass into ethanol in one step, but is not used at the industrial scale yet because the first step, breaking down the plant's cellulose, is much too inefficient. The key, Wu surmised, is to find out what enzymes the bacterium uses to accomplish its feat, and then boost its ability to produce those enzymes. The problem, however, lies in the fact that C. thermocellum uses more than 100 enzymes, and any of the millions of combinations of them may be the magic mixture to break down a particular biomass.

So, Wu decided to make the bacterium do the work for him.

"The bacteria know how to express just the right genes to break down any particular biomass substrate, and we wanted to know how they know to turn on and off just the right genes at the right time to do the trick," says Wu. "We found the bacterium essentially throws the whole bowl of spaghetti at the wall, sees what sticks, and then makes a lot of that particular noodle."

C. thermocelllum produces low levels of many of its enzymes at any one time. When the bacterium comes in contact with wood, for instance, a few of its enzymes break down some of that wood. A product of that tiny reaction is a sugar called laminaribiose that diffuses into the cell. There it deactivates a repressor for two genes, which wake up and start pumping out the two triggers the full production of wood-degrading enzymes CelC and LicA.

Wu's paper shows the first time the triggering pathway for enzyme production in this bacterium has been revealed, and it was only possible because C. thermocellum genome was just recently sequenced, thanks to Wu's collaboration with the U. S. Department of Energy. With its 100 busy enzymes, the entire genome had to be observed as a whole, since fiddling with combinations of two, three, or more enzymes at a time would have taken "more than our lifetime," Wu says.

Wu is now working to re-engineer C. thermocellum to express an abundance of particular genes so it can readily and efficiently produce ethanol from a particular biomass. He's also continuing the genome-wide search for enzyme combinations that will degrade and ferment grasses, corn stovers, and even food waste.

"I don't think this is the revolution that makes ethanol a mainstay," says Wu, "but I believe this is a part of what will lead to the revolution."

Marvel Announces New Spider-Man Cartoon!

Spider-Man swings back into television action in early 2008 as an animated series from Culver Entertainment to air on Kids’ WB! on The CW, it was announced by Kids’ WB! Senior Vice President and General Manager Betsy McGowen and Sony Pictures Television Co-President Zack Van Amburg.

Tentatively titled “The Amazing Spider-Man,” the new series is being produced by Culver Entertainment. Greg Weisman (”Gargoyles,” “The Batman”) is supervising producer, Victor Cook (”Hellboy: Blood and Iron” and “Buzz Lightyear of Star Command”) is producer/supervising director. The series will premiere on Kids’ WB! on The CW, the No.1 rated Saturday morning kids broadcast slate.

Volvo XC70 – 2008 model premiers in Geneva

Simultaneously with the new V70, Volvo’s unveiling its crossover sibling, the 2008 XC70 at the 2007 Geneva Show. Like its predecessor, the XC70 is basically a higher V70 (+75mm) with 4x4 design elements like the scratchproof protective cladding around the wheelarches and the chunkier, redesigned bumpers. Available in Europe from this fall (US should get it a few months later), the XC70 will be offered with two motors, a 238Hp 3.2-litre straight six-cylinder petrol and a 185Hp 2.4-litre five-cylinder D5 turbo diesel. Available with Geartronic transmission only, the 3.2 accelerates from 0 to 60mph (96km/h) in 8.1 sec and achieves a 215km/h - 134mph top speed. The D5 is offered with a 6-speed manual gearbox or a six-speed Geartronic transmission. With the manual “tranny” the D5 accelerates from zero to 60 mph in 8.8 seconds reaching a maximum speed of 130 mph – 209 km/h.

Created in conjunction with fabric designer and manufacture JAB Anstoetz, Chrysler’s Crossfire study attempts to export material and textile styles from the world of… carpets to the world of cars. And if the exterior color scheme doesn't raise an eyelid, without doubt, the light-blue and chocolate-brown toned interior that combines suede and crocodile-leather is something you don't see. Well, that's not entirely true as to be honest, I can think of a chap or two who've decorated their cars "Anstoetz" style. Only they don't wear Armani costumes and they still think that mullets are cool…

Artega GT: VW 3.2 V6 FSI powered sports car premieres in Geneva

Unlike its exotic appearance suggests, the Artega GT which will debut in a couple of days at the Swiss Show is a production model that’ll be manufactured in a limited series of 500 units per year. Its designed by Henry Fisker who’s not only responsible for many of the current Aston Martins like the DB9 Volante but also as most of you remember, for the BMW 645i and Mercedes AMG SL55 based Latigo CS & Tramonto concepts.

2008 Mazda2 – Mazda Demio official image gallery breaks cover prior to Geneva debut


Well, to be honest, not much of a leak this time as only a couple of days remain before the all-new Mazda2 aka Mazda Demio in Japan, debuts at the 77th Geneva Motor Show. Unlike the current model whose conservative looks (unsuccessfully) balance between a semi-mini-mpv and a compact hatch, the new Mazda2 / Demio takes on a modern, you could even say a sporty approach to the mini-segment. That’s even more obvious in the versions fitted with the “Sports Appearance Package”. As you can see by the grey colored car in the pics, this pack adds a new front bumper with a wider and opposite angled air intake, a different grille, side skirts, a roof spoiler and larger alloys. Personally I like it however, I can’t help seeing some Toyota Yaris in it, but maybe that’s just me.

Audi RS6 ABT Crash – Yes, he made it out alive without barely a scratch!

First of all, what you’re seeing in the image above is ONE, and I repeat ONE single car! Makes our previous post on the BMW M3 CSL accident look like foreplay doesn't it? According to the guy who posted these pics on “RS6”, the driver of this split-up Audi RS6 ABT is his brother-in-law. We’re talking about one hell of a lucky chap here as he obviously passed through the valley of death and took a wink at the devil to come out of this crash alive with barely a scratch or two!

Holden HSV GTS gets... "Hot Wheels"

This special edition of Holden’s latest HSV GTS (basically the same model as the HSV Clubsport R8 / Vauxhall VXR8) dressed in a "Hot Wheels" suite was presented at the Melbourne Auto Show. Not bad but we would’ve liked to see this HSV wearing a different set of "hot wheels", preferably black or dark grey colored rims.

Koenigsegg CCGT race car & 1018Hp Bio-Fuel Powered CCXR to premier in Geneva

Koenigsegg’s going to present two new models next week at the Geneva Show. Starting off with the homologated GT1 racer, the CCGT, in order to comply with the ACO and FIA GT1 regulations the Swedish supercar maker removed the superchargers from the stock 806Hp 4.7 V8 and bored it out to 5.0-liters. As for the CCXR., looks like the “environmental friendliness” craze hit those Swedish fellas. The CCXR is equipped with an updated version of the 806Hp 4.7 V8 engine of the CCX that runs on E85 (bio-fuel). Funny enough, due to the fact that the biofuel has higher octane and cooling characteristics, the power has gone up to 1018 hp at 7200 rpm and the torque to 1060 nm at 6100 rpm!

Start your engines: Nissan’s Geneva stand to host PlayStation 3 equipped with Gran Turismo HD

I’m more than willing to place a bet with anyone that more people will be carried away with the new Playstation 3 and the Grand Turismo HD concept game than with the new X-Trail at Nissan’s Geneva stand. We remind you that the Playstation 3 hasn’t gone on sale yet in Europe so this will be a first encounter for most with Sony’s new game console. As for the Gran Turismo HD, it’ll give us a glimpse of Sony's long awaited next gen racing game. Players start with one car on the Eiger Nordwand track in the Swiss Alps. If the track is completed within a restricted time limit, another car is unlocked. There are nine further cars to unlock in this way.

Italdesign Giugiaro Vadho: BMW powered Geneva Concept

Along with their L.A. Show stopper, the Mustang Concept that’ll make its Euro debut, Italdesign and Giugiaro will premier a brand new sports car concept that goes by the name, Vadho at next week’s Geneva Show. This tandem two-seater has borrowed BMW Hydrogen 7’s dual powered V12 (it runs on gasoline and hydrogen) that has an output of 260Hp. The V12 is positioned on the center right of the car next to the passengers. I know this is a bit off topic but the Vadho brought to my mind another Italedesign BMW-powered concept, the 1991 Nazca C2 and its 1993 Spyder sibling. And you know something; 16 years after, the Nazca still makes my heart beat like a Bavarian V10. What a shame that BMW never considered putting this exquisite piece of art into production…

Sbarro Cruise Crosser to debut in Geneva

A Geneva Show without a few weird concepts from Frank Sbarro is like eating a hot dog without ketchup & mustard. We’re not quite sure yet though if the Swiss based coachbuilder will introduce another concept apart from this 6wheeler, pick-up version of the Mitsubishi Outlander based Citroen C-Crosser (we were very specific here, weren’t we…).

Scoop: 2009 Volvo S40

Taking several design cues from the XC60 SUV concept, German magazine Autozeitung has come up with a photoshoped image of the next generation S40 which is expected to come out late 2008 - early 2009. Although nothing’s certain yet, according to a previous report I read on Automotive News who citied a Volvo manager, the 2009 S40 will replace both S40 and S60 models. If that happens, inevitably, the new S40 will grow in length to over 4.5m (matching the BMW 3-Series, Mercedes C-Class & Audi A4) thus closing the gap on today's 4.58m long S60. Fender-Bender Cost Estimates: Unless you own a '81 Escort be prepared to "bend" your pockets

The Insurance Institute for Highway Safety, a nonprofit group funded by the nation's insurers, has released cost estimates for minor fender-bender accidents at low speeds that happen every-day all over the world. Only 3 midsize cars among 17 the Institute tested — the Mitsubishi Galant, Toyota Camry, and Mazda 6 — withstood 4 bumper tests with $1,500 damage or less in each test. Some cars sustained more than $4,500 damage in just 1 of the 4 tests, and 2 cars rang up more than $9,000 total damage. The Volkswagen Passat had the most expensive repair bill for a full front low speed collision at $4,594, followed by the Pontiac G6 which cost $4,588 to fix. But here’s the kicker- The Institute also performed some low-speed crash tests on an older vehicle for comparison. The 1981 Ford Escort aced the 3 mph corner tests with zero damage and had, by far, the lowest repair costs with front corner and rear corner crashes producing no damage. Costs to repair the front accident was only $86, while the rear collision repair bill came to $383

Turns out though that the Russian love affair between Volga’s and BMW’s has been running on quite a few years as this customized Volga 21 RCM proves. Ouchhh... First public Bugatti Veyron crash


As our fellow bloggers on Autoblog.com correctly note, the only other time that we’ve heard about a crashed Veyron was during the development tests. Actually that happened twice resulting in bad publicity and a significant delay of the presentation date. Coulter Oblivious to Damage Done

Two days after Ann Coulter’s CPAC f-bomb and the responses continue to accumulate. Bryan Preston, now that he’s not running around the conference doing his Hot Air gig, gets to the damage Coulter’s words have:

There’s a great deal of political silliness and wishful thinking in that letter–it is a political money pitch for a third-place candidate, after all–but at its core it contains a message that’s likely to resonate. That message is, Republicans are just hateful, horrible people and you need to stop them (by giving us your money, now for the campaign, and in the future through taxes). True, Democrats always say that. But Miss Ann gave them a shred of evidence upon which to convict us all.

And, what’s the big headline coming out of CPAC? Is it that Josh Sparling won a much-deserved award? Is it that conservatives remain committed to the war for freedom against terrorism? Is it that this post-blowout CPAC was the largest CPAC ever? Was it that I got to be on a teevee show?

No. The headline coming out of CPAC is that Ann Coulter said an awful thing. Which is what she wants, since it’ll keep her profile up and help her sell books.

“It’s all about Ann. And that’s the problem,” he concludes.

Fausta, a credentialed CPAC weblogger like myself, decries the intellectual damage from Coulter:

Weak thinking is contagious because it’s easy. It’s a heck of a lot easier than looking at the world and trying to find out what is going on, and what the issues are.

One aspect of CPAC is all the college kids running around. Many idolize Coulter. Her lack of intellectual substance sets a poor example for those people to take back to their schools.

What was Coulter’s response to the anger and disappointment? Pure flippancy:

C’mon, it was a joke. I would never insult gays by suggesting that they are like John Edwards. That would be mean.

Contrasting Approaches to Dealing with Ann Coulter

Kirsten Powers and Lorie Byrd take two different tacks with Ann Coulter and her verbal bomb throwing. Powers takes the “ignore Ann” approach because writing and talking about her simply feeds her ego (about the only thing she eats by the look of things):

I have long been an advocate of ignoring her, but nobody seems willing to take me up on that idea. Can you imagine the epic depression that would engulf her if she made one of her pre-fab attacks and all she got was a collective yawn? What happens to the caricature when the audience disappears? A few cackles at a conservative conference isn’t going to satiate whatever drives her — the drive that forces her to hike up her skirt, drape her cleavage with a cross and insist repeatedly that she is a devout Christian as she spews her famous brand of hateful rhetoric. No, she needs the world to react to her.

I understand her point, but Coulter’s “jokes” are damaging to the conservative movement. I think it’s too important not to speak out. The movement is at the point where her bad, stand-up routine turns more people away than it brings in. I’m not going to let a scrawny, blond gasbag hijack a proud, thoughtful political tradition for a few yucks.

So I lean to Byrd with her post “Ann Coulter Doesn’t Speak For Me:”

I have cheered Ann Coulter in the past when I have agreed with her. I have also spoken out when I have disagreed with her. Any conservative who does not agree with her recent comments should speak out now, as loudly and forcefully as possible. Ann Coulter knows the power of words and she knows how closely all of her words are scrutinized. Those of us who have any kind of forum to speak on behalf of the conservative movement need to realize that our agreement, disagreement or silence will be scrutinized as well.

Conservatives are better than Coulter. It behooves us to speak out. If not we allow Coulter’s awful rhetoric to be conservatism’s voice while damaging our own credibility.

Giuliani Popular with Conservatives Without Even Trying

Mitt Romney and Rudy Giuliani can both claim victory at the CPAC straw poll, but Giuliani was the true winner. Romney got 21% of first choice votes. Rudy was the consensus second choice. When the first and second choices were combined Rudy led with 34% with Romney behind at 30%.

Mitt’s victory came from a very organized campaign. Students were bussed into hand out stickers, hold signs, shout loudly, and clean up Romney flip-flops strewn throughout the exhibit hall.

Giuliani in contrast simply swept into CPAC, gave a speech, and swept out. He was the Mazda candidate going “zoom zoom.” Rudy didn’t have a organization working yet he did very well in the straw poll.

Much of that support comes from conservatives wanting to win the Islamist War. 30% of voters thought the Iraq War was the reason for the GOP electoral defeat last fall. These conservatives think a strong leader winning the war would lead to further success at the ballot box.

Conventional wisdom says the conservative movement is one-half fiscal conservatives and one-half social conservatives. Passionate conservatives that attend CPAC don’t fit into that box. CPAC voters were 50% fiscal, only 30% social, and 19% homeland security/national defense. That may also explain some of Rudy’s strong showing. Sen. Sam Brownback won the social conservatives with Romney and Giuliani splitting the fiscal conservatives.

McCain dissing CPAC obviously hurt him. Only 12% of voters made him their first choice. Newt Gingrich beat him with 14%, and he isn’t even running–yet.

Without even trying Rudy Giuliani wowed a group on paper he has no business wowing. We see Romney has a good, effective organization, but Giuliani is in a good place. With a strong organization he could run away with the nomination.
